I am having the same issue which started immediately after installing 10.6.1 through Software Update. I was having zero issues with sound in 10.6. In 10.6.1, when I go into System Preferences-->Sound, the "Select a device for sound output:" list shows "No output devices found".
For the record, I am using an early 2009 MacBook Pro. I have already reset the PRAM and I actually received the Mac startup sound right after the reset. However, the Mac startup sound is not audible in subsequent restarts. Any clue if this is a driver glitch with a particular subset of Mac hardware?

10.6.1, or 10.6?
10.6.1 is an update to the OS, and still under development, and is only available to ADC members, under a non-disclosure agreement. 10.6 is the one in the shops right now. Is the Mute button ticked in your System Preferences > Sound? |

10.6.1 was released via the software update yesterday.
I have the same problem - no system sounds or for Apple apps (such as Mail, ichat, etc). All other sounds are fine (Skype, etc). Interestingly, the sound device is shown in the preference panel and is working (or I would not be able to hear Skype sounds). It just affects system sounds. I have tried 32 bit and 64 bit kernels, rebooting, permissions, PRAM reset, etc. Very odd. |

I was having exactly the same problem - very annoying.
The files are located in: Macintosh HD > Library > Preferences > Audio OS X should create new ones but it might be a good idea to back them up. Select the plist files in the Audio folder and delete them. Then restart and hopefully that will resolve the problem. Screen recording video on youtube: http://www.youtube.com/watch?v=IPtr0xxqfKc
